Sprite sheet tutorial?

I'm wondering if anyone out there knows an efficient way to create large sprite sheets, or if there's a program that does it for you (preferably not browser-based). I decided to create my own Fallout sprite sheets for use in sprite comics, but I'm having trouble getting the sheets actually put together -- doing it manually is incredibly time-consuming and tedious, and the one program I've tried (Sprite Sheet Packer, it's called) doesn't seem to want to do large sheets.

You can try Hoeloe's program here. I've never used it myself but I think it does what you want.

If not, then you might have to do it by hand. If you know how you would want it to be automated then I could maybe make a program specifically for this though, if it isn't too hard.

Unfortunately Hoeloe's program doesn't support .gif format. It does look promising, though!
