After reading through the spriting dictionary, try to start by making something simple and easy to make, like say a goomba or a Kirby sprite for example.
As for software, I prefer to use ms paint as that is all I really need. Ms paint lacks transparency and layers though, so Graphics gale would be another option as it contains these functions. Really any paint program would work.
