From one of the recent IGN reports, they say they beat the normal mode in an hour. They didn't get to try Last Stand or w/e it's called.
We can hope for a difficulty change option.

Title Defense mode is essentially a hard mode option. The IGN podcast you're referencing mentioned (I think. I only listened to it once) that King Hippo's belly bandage is gone, so the puzzles will be new too. They said the game was pretty easy up to the last quarter. But, let's be honest here: So was the NES game.
